- Active against intra-erythrocytic Plasmodium, by binding haematin in the parasite digestive vacuole
- Retinal toxicity at 100 g total ingestion
- So if on weekly prophylaxis for 5 years, they should get twice-annual retinal screening
- Risk of seizures, psychosis, and worsening psoriasis
